Data Acquisition and Analysis

Samples from the ship's non-toxic supply were collected along the cruise track.

Underway surface sampling was conducted ~2 hours from the non-toxic sea water supply (on occasion at sampling stations surface water was collected with a bucket) and 0.5-4 litres was filtered through 25 mm GF/F filters. Pigment samples were stored at -80 o C until analysis by HPLC which was conducted on board the James Clarke Ross. Pigments were extracted in acetone using ultrasonication, and centrifugation to remove debris, and analysed using the method of Barlow et al. (1997) on a Schimadzu HPLC coupled to a Thermo Separations Products autosampler and a UV6000 diode array absorbance detector. Chlorophyll and carotenoid standards were obtained from Sigma Chemical Co, UK, VKI Water Quality Institute, Denmark and Mike Ondrusek, university of Hawaii.

BODC Data Processing Procedures

Data were submitted to BODC in Microsoft Excel spreadsheet format and saved to the BODC archive with reference PML020057. The file was provided to BODC with sample number, date, sampling position (LAT and LON) and depth as metadata. There were additional non-toxic supply sample data provided in the file with the HPLC pigment data collected by CTD bottle. All the non-toxic sample data were loaded together to check for duplication. Two samples were present in both files and only one was loaded in each case. Sample metadata were checked against information held in the database. The sample number did not match the system recorded in the cruise report and samples were matched based on date and position - aside from slight differences in lat and lon there were no discrepancies.

The data were provided in units of micrograms per litre. The units associated with the parameter codes in the BODC Parameter Dictionary were nanograms per litre, with the exception of chlorophyll-a which was in units of milligrams per cubic metre. All data had a unit conversion factor applied (x 1000) except the chl-a data where the units are equivalent.

Parameter codes defined in BODC parameter dictionary were assigned to the variables as shown in the table below. One additional parameter in the file was not loaded to the database: "TChla" was derived as the sum of "DVChla" and "Chla" and so was not uploaded to the database.
