Perkin Elmer 2400 Series II CHNSO Elemental Analyser
Basic Information
The PerkinElmer 2400 Series II CHNS/O Elemental Analyzer (2400 Series II) is an instrument for the determination of carbon, hydrogen, nitrogen, sulfur or oxygen content in organic and other types of materials. It has the capability of handling a wide variety of sample types.
The 2400 Series II CHNS/O Elemental Analyzer is capable of operating in three modes: CHN, CHNS and Oxygen, is fully automated, and includes a 60 position autosampler. The analyser has an accuracy of <=0.3% and a precision of <=02%. The analytical range is from 0.001 -3.6 mgs for C, 0.001-1.0 for H, 0.001-6 mgs for N and 0.001 to 2.0 mgs for S and O.

Non-toxic (underway) sea water supply
A source of uncontaminated near-surface (commonly 3 to 7 m) seawater pumped continuously to shipboard laboratories on research vessels. There is typically a temperature sensor near the intake (known as the hull temperature) to provide measurements that are as close as possible to the ambient water temperature. The flow from the supply is typically directed through continuously logged sensors such as a thermosalinograph and a fluorometer. Water samples are often collected from the non-toxic supply. The system is also referred to as the underway supply.

Originator's Protocol for Data Acquisition and Analysis
Introduction
The RRS James Clark Ross cruise JR16001 was undertaken as AMT 26 as part of the Oceans 2025 project in a transect across the Atlantic Ocean between 20 September 2016 to 05 November 2016.
A series of CTD and underway samples were collected for analysis of particulate organic carbon (POC), particulate inorganic carbon (PIC), coccolith enumeration (cell counts) and biogenic silica concentration (BSi). The purpose of these samples was to provide an assessment of the inorganic and organic particle concentrations in surface water, provide indices of community composition, and analytical means to calibrate satellite PIC algorithms.
Underway Sampling methodology
Discrete underway samples were collected from the ship's Surf-Met (underway surface and meteorological data collection) flow system in the prep lab 3 to 4 times per day.
Analytical methodology
PIC samples were collected on 0.4 μm polycarbonate filters, rinsed with potassium tetraborate buffer, dried and stored in metal free centrifuge tubes. These were analysed by ICP-OES for particulate calcium.
Coccolith and cell counts were collected on 0.45 μm Millipore HA (nitrocellulose) filters, rinsed with potassium tetraborate buffer, frozen at 20°C, dried, then mounted onto slides using Norland Optical Adhesive and enumerated by birefringence microscopy.
Biogenic silica (BSi) samples were filtered onto 0.4 μm polycarbonate filters, dried in clean centrifuge tubes, and will be analysed following the protocol of Brzezinski and Nelson (1989).
POC samples were filtered onto pre-combusted glass fibre filters, rinsed with 0.2 μm filtered seawater, dried and subsequently fumed with concentrated HCl to remove inorganic carbon. They were analysed post-cruise at Bigelow Analytical Services.
Blanks for filtered samples were collected twice weekly.
