PurposeResearch
ObjectivesAMT20 (JC053) is the third cruise of the third phase of the Atlantic Meridional Transect (AMT) programme. The programme is hosted by Plymouth Marine Laboratory in collaboration with the National Oceanography Centre, Southampton, provides an exceptional opportunity for nationally and internationally driven collaborative research, and provides a platform for excellent multi-disciplinary oceanographic research. As an in situ observation system, AMT informs on changes in biodiversity and function of the Atlantic ecosystem during this period of rapid change to our climate and biosphere.

The aims of the AMT programme [www.amt-uk.org] are to quantify the nature and causes of ecological and biogeochemical variability in the planktonic ecosystems of the Atlantic Ocean, and to assess the effects of this variability on biological carbon cycling and air-sea exchange of radiatively active gases and aerosols. Between 1995 and 2005 marine and atmospheric data were collected twice a year along a 13,500 km transect in the Atlantic Ocean. The cruise track enabled biogeochemical measurements to be made within the poorly studied North and South Atlantic oligotrophic gyres as well as within equatorial and coastal upwelling regions. The range of ecosystems sampled has facilitated the calibration and validation of newly developed techniques, provided a testbed for comparative ecology and enabled the development of atmospheric and oceanographic models. The unique AMT dataset continues to be deposited and made available to the wider community through the British Oceanographic Data Centre.

The programme has provided a platform for national and international collaboration (~80 scientists from 11 countries), and has contributed to the training of >70 PhD students and staff. AMT has produced >170 peer reviewed publications.

Description: Vertical profiles from CTD Niskin bottles at stations along the cruise track before dawn and at solar noon (total CTD stations- 66). 5 depths sampled per station from depths corresponding to the nominal depths of 97%, 55%, 33%, 14% and 1%. 10L of seawater from these depths were filtered via a 0.2 um (47 mm) Millipore nitrocelulose membrane filters for DNA extraction and analysis of coccolithophores and associated viruses. Additional 20L were taken from 97% and 33% and filtered for total lipids analysis via a 1.2 um GF/C filter. All samples were snap- frozen and stored in -80C for further analysis. 1 ml of each sampled depth was also stored in 10 ul of glutaraldehyde for Flow Cytometry analysis. 50 ml from each depth were also syringe filtered via a 0.2 um filter for the isolation of free viruses into a 50 ml falcon tube and stored at 4C for further analysis.

Description: Vertical profiles from solar noon CTD Niskin bottles at stations along the cruise track. Water samples taken at average six depths per station (300 m, 200 m, 100 m, DCM, bottom of the mixed layer, middle of the mixed layer and the water surface, were filtered through the 0.2 micrometre filters. Filtered water was used for spectrophotometric scans for measurements of the CDOM absorption with use of the liquid capillary wave guide cell system - LWCC and the point source integrating cavity absorption meter - PSICAM. Subsamples of the filtered water were use for measurements of the fluorescence Excitation Emission Matrix spectra with use of the Varian Cary Eclipse spectrofluorometer.
