ObjectivesD369 was the last of three National Oceanography Centre 'process study' research cruises to be run by the Ocean Biogeochemistry and Ecosystems research group under the NERC Ocean 2025 research programme. The cruise was aimed at studying effect of ubiquitous eddies on links between microbial carbon, nitrogen and phosphorus cycling (LINK) in the North Atlantic subtropical gyre. The repeated alternating SeaSoar and CTD surveys were aimed at observing physical and biogeochemical changes in eddies and adjoined waters. Daily satellite images and in-situ vessel mounted acoustic current profiling were used to monitor the movement of eddies. The effect of eddy dynamics on microbial community composition, spatial distribution and functioning in the photic layer and twilight zone were studied under the follow objectives:
1. Seasoar and CTD surveys were used to monitor water movement with the aim to estimate mesoscale nutrient fluxes into the photic layer.
2. Vertical mixing in the photic layer was assessed using a turbulence profiler.
3. Nutrients at both nanomolar (nitrate and phosphate) and micromolar concentrations were measured in CTD bottle samples.
4. Coarse-scale (CTD bottle sampling) and fine-scale (PumpCast sampling) vertical distributions of microorganisms were determined for dominant microbial prokaryotic and eukaryotic groups, analysed by flow cytometry. Concentrations of larger microplankton organisms were assessed using the size-fractionating net hauls and FlowCam instrument.
5. Microbial uptake of nutrients were were quantified using radioactive and stable isotope tracers. Group-specific uptake of bicarbonate, phosphate and different nitrogen compounds were determined and corresponding group-specific rates were assessed using flow sorting.
6. Pelagra sediment traps were deployed to estimate biogenic sedimentation in the twilight zone. CTD samples for particulate calcite, opal, and POM measurements were collected.
7. Deep-water incubator was technically tested for measuring microbial respiration in the twilight zone.
